清开灵有效组分调控大鼠脑MVEC缺血再灌注损伤MMP-9mRNA表达的研究

摘    要:目的:观察大鼠脑微血管内皮细胞缺血再灌注损伤MMP-9表达的影响,探讨清开灵有效组分保护血脑屏障阻抑脑缺血再灌注损伤的作用途径。方法:体外培养大鼠脑微血管内皮细胞,氧糖剥夺法模拟缺血再灌注损伤,采用RT-PCR方法半定量检测MMP-9mRNA的表达。结果:与正常组比较,模型组MMP-9mRNA表达明显增加(P<0.01);与模型组比,黄芩苷及新清开灵复方显著降低MMP-9mRNA的表达(P<0.05)。结论:清开灵有效组分可能通过减少血脑屏障内皮细胞MMP-9mRNA表达而阻抑脑缺血损伤炎症反应。

Abstract:Objective:To observe the expression of matrix metalloproteinase-9 mRNA of cultured rat brain microvascular endothelial cells(MVEC),expecting to explore the mechanisms of QingKaiLing effective components protecting brain from injury of inflammatory cascade in cerebral ischemia diseases.Method:Primary culture of rat brain MVEC,establishing an in vitro ischemia reperfusion model(OGD),using RT-PCR method to detect the expression of MMP-9 mRNA.Result:The expression of MMP-9mRNA of model group are significantly higher than normal group(P<0.01),Baicalin and new QingKaiLing injection can depress the expression(P<0.05).Conclusion:The function of QingKaiLing effective components on inflammatory cascade in cerebral ischemial diseases is probably by intervention of the expression of MMP-9.
