高糖加高胰岛素对心肌成纤维细胞增殖的影响

摘    要:目的 利用体外培养的乳鼠心肌成纤维细胞,观察高浓度葡萄糖加高浓度胰岛素对心肌成纤维细胞增殖的影响,并探讨其可能作用机制。方法 以培养的乳鼠心肌成纤维细胞为模型分组给药后,利用结晶紫染色法观察细胞的增殖情况;利用^3H]thymidine标记法测定细胞DNA的合成;利用流式细胞仪分析细胞周期。结果与正常对照组相比,高糖、高胰岛素、高糖加高胰岛素组分别造成了心肌成纤维细胞数目、DNA合成及S+G2+M期细胞的百分比增加,(P〈0.01),而高糖加高胰岛素组的各项指标值显著高于其它各组,(P〈0.01)。结论 高糖加高胰岛素具有协同促进心肌成纤维细胞增殖的作用。

Abstract:Objective To study the effect of high glucose and high insulin on cardiac fibroblast proliferation. Methods The neonatal rat cardiac fibroblasts were cultured and treated with different factors.Cell proliferation was measured with cell number and the DNA synthesis assayed with thymidine intake method,the percentage of S+G_2+M in cell cycle. Results After being treated with high glucose,high insulin,high glucose and high insulin,the cell number,DNA synthesis,the percentage of S+G_2+M in cell cycle were increased significantly(P<0.01) compared with control group.High glucose and high insulin group showed a further increase compared with high glucose group and high insulin group(P<0.01).Conclusions High glucose and high insulin can coordinately promote proliferation of cardiac fibroblasts.
