胶质瘤端粒酶活性与瘤增殖能力的关系研究

摘    要:目的:通过对胶质瘤端粒酶活性的测定和Ki-67核抗原免疫组化染色,了解端粒酶活性与胶质瘤预后的增殖潜能之间的关系。方法:对42例胶质瘤组织标本,用端粒酶PCR ELISA盒进行端粒酶活性半定量一,免疫组织化学染色检查Ki-67抗原。比较各组肿瘤端粒酶活性,Ki-67表达的差异,结果:正常人脑血组织无端粒酶活性。胶质细胞瘤Ⅱ级组1/15例(6.7%),Ⅲ级组5/13(38.5%)、Ⅳ级组10/14例(71.4%)端粒酶阳性,各组之间阳性率有显著差异,Ki-67阳性细胞表达指数则随肿瘤恶性程度增高而增加结论:端粒酶在胶质瘤的发展中起重要作用,它是脑肿瘤发展过程中,晚期细胞分裂旺盛的分子标志。

Abstract:Through detecting the telomerase activity of brain gliomas and combing with ki-67 immunohistochemical staining analysis, we are trying to reveal the relationships between the telomerase activity and the tumorigenesis, development, and proliferation of gliomas. Methods: Telomerase activity in glioma tissues from 42 patients with glioma were detected by telomerase PCR and ELISA. Ki-67 antigen of the specimen was examined by immunochemical stains. Results: Normal adult brain tissues had no detectable telomeras activity. Whereas, 1/15, 5/13, and 10/14 cases of gliomas grade II, III, IV had detectable telomerase activity (6.7% , 38.5% and 71.4% respectively). The positive rate of telomeras activity was significant higher in high-grade glioma. Conclusion: Telomerase may play an important role in the development of malignant glioma. The presence of telomerase in tumor may be a marker to predicate the worse prognosis.
