摘    要:用体外短期液体培养技术,观察RA和不同来源的造血调控因子对HL-60细胞增殖与分化的影响。以生长曲线和倍增时间作为观察各种试剂对HL-60细胞增殖作用的指标;以NBT还原反应和细胞形态变化作为分化成熟的指标。结果是RA、PHA-LCM单用时对HL-60细胞均有抑制增殖和诱导分化作用,PWM-SCM、LP3-CM仅部分抑制增殖,而FGF、Hu-CSF和EPO等无效。各种造血生长因子分别与RA合用时,能加强RA对HL-60细胞的增殖抑制和诱导分化作用的有FCF、PWM-SCM、PHA-LCM和Hu-CSF等;LP3-CM仅增强抑制增殖:EPO未显示作用。

Abstract:Using a short-term liquid culture assay, the effects of induction of differentiation of HL-60 cells by retinoic acid (RA) and several hematopoietic modulation factors were evaluated.The results demonstrated that (1) RA and PHA-LCM were capable of inhibiting proliferation, and inducing differentiation of HL-60 cell line, respectively. (2) PWM-SCM and LP3-CM had partial an inhibitory effect on HL-60 cell growth while FGF, Hu-CSF and EPO had no such an effect.(3) although any one of hematopoietic modulation factors by itself had no siginficant effect on induction of differentiation of HL-60 cell line, it could euhance granulocytic maturation when used in combination with RA, except for LP3-CM and EPO.
