手法对兔膝骨性关节炎软骨细胞DNA氧化性损伤的防治作用

摘    要:为研究手法对膝关节骨性关节炎治疗的作用机制,采用新西兰兔进行动物造模而形成膝关节骨性关节炎模型,随机将实验动物分为手法组、施沛特组和对照组并进行实验。8周后取动物右膝关节软骨进行DNA提取,并采用LCMS2010型液-质联用仪检测标本中8-羟基脱氧鸟苷含量。结果显示,施沛特组含量最少,手法组次之,对照组最多。提示手法治疗可能对减轻氧自由基对软骨细胞DNA的损伤有一定的作用。

Abstract:In order to study the action mechanism of the manipulations in the treatment of knee osteoarthritis(KOA),new Zealand rabbits were made into KOA models and randomly divided into three groups: Group A,Group B and Group C.Group A and Group B were treated by the manipulations and by intra-articular injection of Sodium Hyaluronate,respectively and Group C gave no treatment;eight weeks later,the DNA was extracted from the right-knee cartilage and the 8-OHDG contents in the samples were measured by LCMS2010 apparatus.The results showed that the 8-OHDG content of Group B was the lowest,the one of Group A was the second and the one of Group C was the most,suggesting that the manipulations could have a certain function to alleviate the chondrocyte DNA injury by oxygen free radical.
