CO2气腹对人宫颈癌CaSki细胞MMP-9和VEGF表达的影响

摘    要:目的 探讨腹腔镜二氧化碳(CO2)气腹对人宫颈癌CaSki细胞株基质金属蛋白酶-9(matrix metalloproteinase-9,MMP-9)和血管内皮生长因子(vascular endothelial growth factor,VEGF)表达的影响.方法 建立腹腔镜CO2气腹体外模型,将CaSki细胞株分别置于0、7和14mmHg的CO2气腹环境下培养1、2、4和8h.采用RT-PCR和Western blot法分别检测处理后各组CaSki细胞株中MMP-9和VEGF表达情况.结果 RT-PCR和Western blot结果显示,与0 mmHg CO2气腹组相比,7和14 mmHg CO2气腹组CaSki细胞株MMP-9和VEGF的mRNA和蛋白表达显著增加(P<0.05),且随着处理时间的延长MMP-9和VEGF的表达呈上升趋势(P<0.05),处理4h时MMP-9和VEGF的表达量达到高峰;在同一时间点时,7mmHg CO2气腹组的MMP-9和VEGF表达量最高(P<0.05).结论 在一定压力和作用时间范围内,CO2气腹可能是通过上调MMP-9和VEGF的表达,进而促进肿瘤生长.

Abstract:Objective To investigate the effects of carbon dioxide on the expression of matrix metalloprotein- ase -9 ( MMP- 9) and vascular endothelial growth factor(VEGF) in human cervical cancer cell line CaSki. Methods The imitated CO2 pneumoperitoneum was established in vitro. After CaSki ceils were exposed to CO2 pneumoperitoneum at 0,7 or 14 mmHg for 1,2,4,8h, the expressions of MMP -9 and VEGF were exam- ined by RT - PCR and Westem blot. Results Compared with 0 mmHg ('.02 pneumoperitoneum group, the ex- pressions of MMP-9 and VEGF in CaSki were significantly increased( F 〈0.05 ) under 7 and 14mmHg CO2 pneumoperitoneum. The expression of MMP -9 and VEGF in CaSki had an increasing tendency with the expo- sure time prolongation, and after treated 4h, the expressions of MMP -9 and VEGF reached peak. At the same time point,the expression of MMP -9 and VEGF in 7mmHg CO2 pneumoperitoneum group was the highest. Conclusion Under certain pressure and action time range, through upregulation the expression of MMP -9 and VEGF, CO2 pneumoperitoneum can promote tumor growth.
