骨肉瘤中细胞凋亡和细胞增殖的关系

摘    要:目的探讨骨肉瘤组织中细胞凋亡与细胞增殖的关系。方法利用原位末端转移酶法和免疫组织化学技术分别检测骨肉瘤和良性骨肿瘤中凋亡细胞密度和增殖细胞核抗原(PCNA)的表达。结果 4种亚型骨肉瘤组织中凋亡细胞密度均明显低于良性骨肿瘤组织 ,而PCNA阳性细胞密度明显高于良性骨肿瘤组织 ;在 4种亚型骨肉瘤中凋亡细胞密度和PCNA阳性细胞密度呈明显负相关 ,而在良性骨肿瘤中两者呈线性正相关。结论在骨肉瘤的发生中可能普遍存在细胞凋亡机制的紊乱 ,可能以低凋亡高增殖表现为多 ,并认为细胞凋亡检测尚难以作为一种独立的预后判断指标

Abstract:Objective To investigate the relationship between apoptosis and cell proliferation in osteosarcomas. Methods The density of apoptotic cells and expression of proliferating cell nuclear antigen (PCNA) were detected by in situ terminal deoxynucleotide transferase labeling (ISTDTL) and immunohistochemical techniques respectively in 65 osteosarcomas and 19 benign bone tumors. Results The densities of apoptotic cells in 4 subtypes of osteosarcomas were significantly lower than those in benign bone tumors, but the densities of positive PCNA cells in former were significantly higher. An obvious negative correlation was found between densities of apoptotic cells and those of positive PCNA cells in osteosarcomas, while positive correlation existed between densities of both cells in benign bone tumors (r=0.4251). Conclusions The data suggest that disturbance of apoptotic mechanism may be a common event in the genesis of osteosarcoma , high density of proliferative cells and low density of apoptotic cells occur more usually in osteosarcoma tissues. Predictive value of apoptosis as an independent prognostic parameter remains to be established in a larger scale.
