摘 要: | 目的 :分析钙拮抗剂异搏定 (verapamil,Ver)联合应用阿霉素 (adriamycin ,ADM)柔红霉素 (daunorubicin ,DNR)、足叶乙甙 (etoposide,VP1 6 )、高三尖杉酯碱 (homoharringtonine,HHT)和长春花碱 (vinblastineVBL)等抗肿瘤药物对人涎腺粘液表皮样癌高转移细胞 (Mc3)的增效效应 .方法 :采用细胞培养、细胞计数及MTT等方法观察各化疗药物对Mc3细胞的杀伤作用 ,根据药物剂量 -效应曲线、半抑制浓度 (IC50 )、抗肿瘤活性 (RAA)比较各化疗药物单独用药及分别联合非细胞毒性的Ver的抗肿瘤作用及联合用药合并指数 (CI50 ) .结果 :Ver剂量超过 32 0 0 0时 ,对Mc3细胞有生长抑制作用 ,其IC50为 3981 0 .72 ;DNR ,VBL ,VP1 6 ,ADM和HHT等药物单独用药对Mc3细胞均呈现具有一定量效关系的生长抑制作用 .低浓度的DNR ,VBL ,VP1 6 ,ADM和HHT与非细胞毒性剂量的Ver联合应用 ,各用药组均呈现极强的细胞毒性作用 ,CI50 分别 <0 .95 ,其所合用的剂量水平均有显著的协同作用 .结论 :非细胞毒性剂量的Ver明显增强了化疗药物的抗肿瘤活性 ,对人涎腺粘液表皮样癌高转移肿瘤细胞生长抑制有明显的增效效应 .

Synergic anti-tumor effect of combination of verapamil with chemotherapy on highly metastatic human mucoepidermoid carcinoma ( Mc3 ) cells |

Abstract: | AIM: To study the inhibitory effect of adriamycin (ADM), daunorubicin ( DNR), etoposide(VP16), homoharringtonine (HHT) and vinblastine (VBL) on human highly metastatic mucoepidermoid carcinoma( Mc3) cells and the synergic anti tumour effect of verapamil (Ver) with these anti tumour drugs. METHODS: Mc3 cells were exposed to ADM, DNR, VP16, HHT and VBL respectively, also to Ver combined with these drugs at various concentrations. The inhibitory effect and synergic anti tumor effect were detected by microculture tetrazolium colorimetric (MTT) assay and cell counting method. The growth curves of cells with different drugs and dosages were drawn. Then IC 50 and doubling time (DT) of cells were calculated. RESULTS: The 50% inhibition concentration (IC 50 ) value of Ver was 32 000. The nontoxic dose of Ver combined with DNR, ADM, HHT, VBL and VP16 showed an enhanced anti tumour effect which displays a dose dependent manner. And the combination index of 50% inhibition (CI 50 ) was lower than 0.95 respectively. CONCLUSION: Combination of Ver with ADM, DNR, VP16, HHT and VBL has a remarkable synergic anti tumor effect on Mc3 cells. |
