摘    要:目的 观察膀胱出口梗阻(BOO)后膀胱平滑肌细胞碱性成纤维细胞生长因子(bFGF)、转化生长因子(TGFβ-1)表达的变化,探讨生长因子在BOO后膀胱平滑肌细胞继发改变中的作用。方法 应用逆转录-聚合酶链反应(RT-PCR)和免疫组织化学SP法在mRNA和蛋白水平检测bFGF、TGFβ-1在30例BOO及4例非BOO膀胱平滑肌组织中的表达。结果 BOO组与非BOO组膀胱平滑肌组织均有bFGF mRNA表达,BO组表达水平高于非BOO组,P<0.01;TGFβ-1 mRNA在两组未见表达。结论 BOO 后磅胱平滑肌的继发改变与bFGF mRNA的表达有关。

Abstract:Objective To study the expression of basic fibroblast growthfactor and transforming growth factor beta 1 in human bladder smooth muscle in BOO. Methods RT-PCR and immunohistochemical study were carried out in 30 bladder tissue samples acquired from patients with identified BOO of BPH on suprapubic prostatectomy and 4 bladder samples acquired from other bladder operation. Results b-FGF mRNA was expressed in bladder smooth muscle cells in both BOO group and control group. The level of expression in the BOO group was higher than that in the control group(P<0.01). TGFβ-1mRNA wasn't expressed in both groups. Immunohistochemical study indicated that bFGF was expressed in the cytoplasm of bladder smooth muscle cells. Conclusion It was suggested that the expression of bFGF mRNA was correlated with the alteration of bladder smooth muscle due to BOO.
