APP17肽对糖尿病大鼠脑枕叶视区皮层神经元退行性变的影响

摘    要:目的 探讨APP17肽对糖尿病(DM)大鼠脑内视觉传导通路枕叶视区皮层神经元退行性变的影响。方法 用链脲佐菌素(STZ)腹腔注射诱发DM模型,10周后取脑组织行免疫组化染色观察神经元内凋亡因子Bax、Bcl-2、Cyto C的表达。同时取枕叶视区皮层处脑组织作电镜观察。结果 DM组枕叶视区皮层17区内Bax、CytoC阳性反应神经元数目多、染色深,Bcl-2较正常组神经元数目少,APP17肽组Bax、Bcl-2、CytoC的表达均接近正常组。DM组大鼠枕叶视区皮层神经元超微结构出现明显损害,给予APPl7肽后病变有改善。结论 APP17肽可改善DM大鼠枕叶视区皮层神经元的退行性改变。

Abstract:ObjectiveTo investigate the effect of APP17 pept ide on the ultrastructure and expression of apoptosis related protein (Bax, Bcl 2, Cyto C) in the occipital visual cortex in diabetic rat.Methods The experimental rats were divided into normal group (NC group), DM grou p, APP17 peptide treated group (APP group). Diabetic rat model was established b y injection of streptozotocin into peritoneal cavity. In APP17 peptide treated g roup, model rats were injected with APP17 peptide subcutaneously, and their brai n sections were prepared after 10 weeks of survival when water maze test was fin ished. The immunohistochemical staning of these sections were then performed wit h Bcl 2, Bax, Cyto C antibody. The rats in control group were injected with sal ine.ResultsBax, Cyto C positive neurons in the occipital vis ual cortex 17 area were more in DM grouop than in the normal and APP17 treated g roup significantly (P<0.01); Bcl 2 positive neurons had no marked differenc e among the three groups. The ultrastructure of the occipital visual cortex in D M group had abnormal changes, but still contain the integrity in APP17 treated g roup.ConclusionThere exists neurodegenearation in the occipi tal visual cortex of diabetic rat. APP17 peptide can improve the degenerative ch anges.
