滤纸片干血斑在HIV-1 BED-CEIA新发感染检测方法中的应用

摘    要:目的评价滤纸片干血斑在HIV-1 BED-CEIA新发感染检测中洗脱液放置时间对结果判定的影响。方法使用2013年新发感染干血斑国际考核盘进行HIV-1 BED-CEIA检测,检验干血斑考核盘在BED-CEIA新发感染检测结果的稳定性与可重复性;洗脱液放置13 h、17 h、20 h、24 h、32 h后检测,进行随机区组设计方差分析。结果检测稳定性较好,6次检测质控品吸光度值(OD值)与标化吸光度(ODn值)的变异系数(CV)〈15%;国际考核盘干血斑样8份,其不同放置时间5次检测判定为新发感染结果一致,即可重复性好;洗脱液放置13 h、17 h、20 h、24 h、32 h后检测,标化吸光度(ODn值)差异无统计学意义(χ2=0.705 7,P>0.05)。结论在HIV-1 BED-CEIA新发感染检测中,干血斑样本的检测结果具有较好的稳定性与可重复性,因此可以在今后检测工作中,应用干血斑样本进行HIV-1 BED-CEIA新发感染检测。

Abstract:Objective To evaluate effect of eluant placement time of dried blood spot specimens in HIV-1 BED-CEIA as- say detection for new infection on result judgement. Methods Dried blood spot panel were taken to evaluate the stability and repeatability of the detection results by HIV-1 BED-CEIA test. Eluant of dried blood spot were tested at the collection time of 13, 17, 20, 24 and 32 hours and the results compared. Results The coefficient of variation (CV) of dried blood spot panel was less than 15% by counting the OD and ODn values of six runs. Eight specimens showed the same results in five tests. There was no statistical significance in ODn when the detection was carried out after the eluant placed for 13, 17, 20, 24 and 32 hours (Z2=0.705 7,/9〉0.05). Conclusions In HIV-1 BED-CEIA detection for new infection, repeatability and stability of dried blood spot samples are excellent and can be used in HIV-1 BED-CEIA detection for new infection furthei'.
