缺血预适应预防大鼠移植肾排斥反应的实验研究

摘    要:目的 研究大鼠供肾缺血预适应对移植肾排斥反应的影响。方法 采用改良的大鼠肾移植模型,将SD大鼠肾移植给Wistar大鼠作为同种异体移植,Wistar大鼠肾移植给Wistar大鼠作为同品系移植的空白对照。共设3个实验组,观察各组大鼠的移植肾功能、粘附分子、细胞因子、MHC-Ⅱ分子的表达及排斥反应发生的程度。结果 缺血预适应能减少移植肾粘附分子ICAM-1及细胞因子IL-2、TNF—α的表达;并抑制MHC-Ⅱ分子在移植肾内的表达,改善移植肾功能。结论 缺血预适应对移植肾有保护作用,抑制MHC-Ⅱ分子在移植肾内的表达,可明显减轻移植肾排斥反应的程度。

Abstract:Objective To explore the protective effect of ischemic preconditioning on transplanted kidney against rejection. Methods Modified renal transplantation model of rats was used. The kidney of SD rat as allograft was transplanted into a Wistar rat. The kidney of one Wistar rat as isograft was transplanted into another as the control . Experimental rats were divided into 3 groups. The changes of renal function, expressions of adhesion molecule, cytokine and MHC- II , and the degree of acute rejection of the transplanted kidney in each group were observed after pretreated renal transplantation. Results Ischemic preconditioning could reduce the expressions of ICAM-1, IL-2, TNF-a and MHC- II , inhibit the expression of MHC- II and improve the renal function of the transplanted kidney. Conclusion Ischemic preconditioning can greatly alleviate renal rejection and protect the transplanted kidney against rejection.
