脂质体包裹99mTc-HYNIC-Survivin ASODN的鸟嘌呤含量与启动子位置对显像质量的影响

摘    要: 目的 制备鸟嘌呤(G)含量及离启动子远近不同的4种脂质体包裹99mTc-HYNIC-Survivin ASODN对荷人肝癌裸鼠行SPECT显像,探讨其显像质量是否受G含量及启动子位置影响,以找到最佳反义显像的碱基序列,提高基因显像质量。方法 制备两种(A1、A2)G含量不同而离启动子远近相近的16个碱基单链ASODN和两种(A'1、A'2)G含量相同而离启动子远近不同的10个碱基单链ASODN,耦联HYNIC,99mTc标记后,经Cellufine GH-25纯化后脂质体包裹。用此显像剂对荷瘤裸鼠行肿瘤反义显像并通过ROI技术进行分析。结果 肿瘤/对侧肌肉比值(T/NT)A1、A2分别为4.13±0.09 和2.47±0.04(2 h),4.90±0.11和3.75±0.10(4 h);A'1、A'2分别为3.90±0.07和2.09±0.07(2 h),4.41±0.08和2.78±0.10(4 h)。上述各组T/NT值在各时间点差异有统计学意义(P<0.01)。结论 G含量相同,离启动子位置较近,显像质量较好;离启动子位置相近,G含量较低,显像质量较好。
