罗格列酮对糖尿病大鼠心肌纤维化及核因子κB、结缔组织生长因子表达的影响

摘    要:目的探讨罗格列酮对糖尿病大鼠心肌纤维化及心肌核因子κB(NF-κB)、结缔组织生长因子(CTGF)表达的影响。方法链脲佐菌素(STZ)诱导糖尿病大鼠模型。30只大鼠分为3组:正常对照组(n=8)、模型组(n=11)和罗格列酮组(n=11)。罗格列酮组每日予罗格列酮5 mg·kg~(-1)灌胃,其余2组予同体积生理盐水,共12 wk。采用Masson染色检测左室心肌胶原含量、免疫组化法检测心肌NF-κB的表达、Western印迹技术检测心肌CTGF蛋白质表达。结果与正常对照组相比,模型组大鼠左室心肌组织胶原含量明显升高(10±s 3)%vs(16.2±2.4)%,P<0.0l;NF-κB、CTGF蛋白质表达水平亦明显升高,P<0.01。与模型组比较,罗格列酮组大鼠左室心肌组织胶原含量有所下降(16.2±2.4)%vs (12±3)%,P<0.05;NF-κB、CTGF蛋白质表达水平明显减少,P<0.01。结论罗格列酮可抑制糖尿病大鼠心肌纤维化及心肌NF-κB、CTGF的蛋白表达。

Abstract:AIM To evaluate the effects of rosiglitazone on the myocardial fibrosis,nuclear factor-kappa B (NF-κB)and connective tissue growth factor(CTGF)expression in myocardium of diabetic rats.METHODS Diabetic rats were induced through streptozotocin.Thirty male Sprague-Dawley rats were randomly divided into 3 groups:control group(n=8)and diabetic model group(n=11),with peffusion into the stomach of normal saline(same volume as rosiglitazone group);rosiglitazone group(n=11),to be constructed into diabetic model with perfusion into the stomach of rosiglitazone(5mg'kg-(-1))once daily.After 12-week treatment,the collagen content of myocardium was assessed with Masson staining,the activation of NF-κB by immunohisto- chemistry and the protein expression of CTGF through Western blotting.RESULTS By the end of the experiment,the collagen content of the diabetic model group was(16.2±s2.4)%,significantly higher than that of the control group(10±3)%,P<0.01;and the collagen content of rosiglitazone group was(12±3)%,significantly lower than that of the diabetic model group(P<0.05),with furthermore of the NF-κB and CTGF expression significantly lower than those in the diabetic model group(P<0.01).CONCLUSION Rosiglitazone inhibits the protein expression of CTGF and NF-κB,which may play an important role in the process of ameliorating myocardial fibrosis in diabetic rats.
