IGF-1和黄芪丹参治疗骨骼肌钝挫伤的生物力学评价

摘    要:目的观察胰岛素样生长因子-1(IGF-1)和黄芪丹参的治疗对大鼠骨骼肌钝挫伤愈合的影响。方法72只雄性SD大鼠,制作成右侧腓肠肌钝挫伤动物模型,随机分成3组,即IGF-1组(注射外源性IGF-1),中药组(注射黄芪丹参注射液)和自然愈合组。于伤后7、14、21、28、42、56d测双侧腓肠肌的最大颤搐强度、断裂强度和拉伸长度。结果(1)自然愈合组损伤骨骼肌力学指标在伤后8周仍明显低于正常;(2)IGF-1组和中药组改善的速度和程度均明显高于自然愈合组,以IGF-1组尤为显著;(3)IGF-1治疗组修复的损伤骨骼肌,其最大颤搐张力和断裂强度均明显高于健侧。结论(1)急性骨骼肌钝挫伤后,自然愈合组修复质量不佳;(2)IGF-1和黄芪丹参具有促进骨骼肌损伤修复的作用,其中以IGF-1的作用更为显著;(3)IGF-1可能有促进骨骼肌肥大的作用。

Abstract:Purpose To observe the effect of insulin-like growth factor-1 (IGF-1) and HuangqiDanshen on skeletal muscle healing following contusion. Methods A standard contusion model was reproduced at the right gastrocnemius in 72 rats. They were randomly divided into 3 groups, i. e. ,group IGF-1, group Chinese medicine,and group natural healing. The 3 groups were injected respectively with exogenous IGF-1, injection of Huangqidanshen, or nothing for control, At day 7,14,21,28,42,56 after injury, the biomechanical testing was performed to examine maximal tetanic tension, failure load, and failure displacement of injured gastrocnemius and non-injured as normal. Results (1) In group natural healing, the biomechanical parameters of injured muscle were obviously inferior to normal even in 8 weeks after injury; (2) The improvement of those parameters in both groups IGF-1 and Chinese medicine was better than in group control, especially in group IGF-1; (3) the maximal tetanic tension and failure load in group IGF-1 were recovered even greater than that in normal. Conclusions (1) The healing quality of skeletal muscle in group control is poor after acute contusion; (2) The healing of injured muscle can be improved by IGF-1 and HuangqiDanshen, especially with IGF-1; (3) IGF-1 may be contribute to skeletal muscle hypertrophy.
