胃癌及癌前病变组织中MUC6核粘蛋白的表达及其意义

目的　探讨正常胃粘膜、癌前病变组织以及胃癌组织中MUC6核粘蛋白的表达规律 ,及其与患者临床病理因素之间的联系。方法　应用兔抗人MUC6核粘蛋白的抗体和免疫组织化学SP法检测人正常胃粘膜 (n =10 )、癌前病变肠上皮化生粘膜 (n =2 9)和胃癌组织 (n =47)中MUC6核粘蛋白的表达。结果　人正常胃粘膜中胃腺的基底部广泛分布MUC6核粘蛋白 (10 0 % ) ,而肠上皮化生、不典型增生和胃癌组织中的表达阳性率分别为 13 .8%、70 %、44.7%。胃癌组织中MUC6核粘蛋白的表达与胃癌患者的性别、肿瘤的部位和大小、淋巴结转移、肿瘤的分化与浸润、临床分期和Lauren氏分型之间差异无显著性 (P >0 .0 5 )。结论　MUC6基因是组成胃粘膜的粘蛋白基因之一 ,在胃癌前病变和胃癌组织中的表达呈下调趋势 ,但与患者的一些临床病理指标之间不存在关联。

Expression of MUC6 apomucin in the tissues of precancerous lesion and gastric carcinoma and its significance

Objective To explore the expression of MUC6 apomucin in thenormal gastric mucosa, precancerous lesion and gastric carcinoma and the association with clinical pathological factors. Methods The specimens were collected from 10 normal gastric mucosa tissues, 29 intestinal metaplasia tissues as precancerous lesion and 47 gastric cancer tissues respectively. The expression of MUC6 apomucin was detected in all specimens by immunohistochemical SP method with rabbit multiclonal antibody against human MUC6 apomusin. Results MUC6 apomucin was extensively distributed in the basilar part of gastric gland in the normal gastric mucosa (100%), but the expression rates were 13.8%, 70% and 44.7% in the tissues of intestinal metaplasia, atypical hyperplasia and gastric cancer, respectively. No association was found between the expression of MUC6 apomucin in the neoplastic epithelium and patients' sexes, locations and sizes of tumor, lymph node metastases, differentiations of cancerous cells, invasiveness, clinical stages and Lauren's types of the disease. Conclusion MUC6 gene, one of the mucin genes expressed in normal gastric carcinoma, its expression is markedly down-regulated in the precancerous lesion and gastric carcinoma, but not associated with patients' clinical pathological factors.
