摘    要:目的:研究C反应蛋白(CRP)对急性冠脉综合征(ACS)患者外周血树突状细胞(DC)分化成熟的影响。方法:研究人群分为ACS组与正常对照组。分离人外周血单核细胞,用含rhGM-CSF和rhIL-4的完全培养液培养7 d,使其分化为DC后加入CRP孵育。应用流式细胞仪检测DC表型CD80、CD86的表达;混合淋巴反应检测DC对T淋巴细胞的刺激作用;酶联免疫吸附法(ELISA)测定混合淋巴反应上清液中IL-4、IFN-γ水平。结果:与正常对照组比较,ACS组DC表面CD80、CD86的表达明显增高,对T淋巴细胞刺激的能力增强;致炎细胞因子IL-4、IFN-γ分泌增多;且IL-4/IFN-γ比值降低。结论:ACS时CRP可明显促进DC的分化成熟,刺激的免疫类型以细胞免疫为主,即产生免疫偏移。

Effects of C Reactive Protein on Activation of Dendritic Cells in Acute Coronary Syndrome

Abstract:Objective:To investigate the effects of C reactive protein on maturation and immunological function of human dendritic cells in acute coronary syndrome(ACS).Methods:Study population was divided into two groups:ACS group and Control group.Human monocytes were isolated and cultured for 7 days in completed medium containing hFM-CSF and rhIL-4 to obtain the immature DCs.The isolated DCs were incubated with CRP for 48 hours.The expression of CD80 and CD86 were detected by FACS.The stimulating capacity of DCs was determined in allogenic mixed lympho-cyte reaction(MLR) . ELISA was used to detect the level of cytokines ( IL -2 and IFN -γ) in the medium of MLR. Results:Compared with the Control group, CD80 and CD86 on DCs surface was much more expressed in ACS group; the stimulating capacity of DCs in MLR was higher; and the T lymphocytes in MLR produced higher levels of pro - inflammation cytokines (IL- 12 and IFN -γ). Conclusions:CRP not only promoted the maturation and immunological function of human DCs in ACS obviously and also made the Th1/Th2 immune deviation.
