摘    要:目的建立川贝清肺糖浆中蔗糖、果糖、葡萄糖、麦芽糖的含量测定方法。方法采用高效液相色谱-示差折光检测(HPLC-RID)法,色谱柱为Alltechchrom Prevail carbohydrate ES 5u(250mm×4.6mm,5μm),以乙腈-水(75∶25)为流动相,流速1.0mL·min^-1,柱温35℃,示差检测器,检测器温度40℃,进样量10μL。结果在该色谱条件下,四种糖类物质能完全分离;蔗糖在0.7811~7.8113mg·mL^-1浓度范围内线性关系良好(R^2=0.9997),平均回收率为99.17%(n=6);果糖在0.5065~5.0657mg·mL^-1浓度范围内线性关系良好(R^2=0.9999),平均回收率为97.84%(n=6);葡萄糖在0.5078~5.0789mg·mL^-1浓度范围内线性关系良好(R^2=0.9999),平均回收率为97.15%(n=6);麦芽糖在0.2501~2.5017mg·mL^-1浓度范围内线性关系良好(R^2=0.9991),平均回收率为94.48%(n=6)。结论本方法专属性高、重现性好,可真实反映川贝清肺糖浆中蔗糖的实际添加情况。

Abstract:OBJECTIVE To establish a method for the determination of sucrose,fructose,glucose and maltose in ChuanBei QingFei syrup.METHODS HPLC-RID was adopted on aAlltechchrom Prevail carbohydrate ES 5u column(250mm×4.6mm,5μm)with mobile phase system acetonitrile-water(75∶25).The flow rate was 1.0mL·min^-1,and the column temperature was maintained at 35℃,RID detector temperature was maintained at 40℃,and injection volume was 10μL.RESULTS Under this chromatographic condition,the four saccharides can be completely separated.The linear relationship of sucrose was good in the concentration range of 0.7811-7.8113mg·mL^-1(R^2=0.9997),and the average recovery rate was 99.17%(n=6).The linear relationship of fructose was good in the concentration range of 0.5065-5.0657mg·mL^-1(R^2=0.9999),and the average recovery rate was 97.84%(n=6).The linear relationship of glucose in the concentration range of 0.5078-5.0789mg·mL^-1 is good(R^2=0.9999),and the average recovery rate is 97.15%(n=6).Maltose has a good linear relationship in the concentration range of 0.2501-2.5017mg·mL^-1(R^2=0.9991),and the average recovery rate is 94.48%(n=6).CONCLUSION The method has high specificity and good reproducibility,and can truly reflect the actual addition of sucrose in ChuanBei Qingfei Syrup.
