Expression of miR-146a in patients with pre-eclampsia and their clinical significance

Objective To explore the expression difference of miR-146 a in the placenta tissues of normal parturients and preeclampsia(PE) patients and its influence on the pathogenesis of preeclampsia.Methods A total of 24 PE patients who were admitted to Linyi Traditional Chinese Medicine Hospital from October 2019 to September 2020 were selected as PE group, and 24 healthy pregnant women who gave birth in this hospital during the same period were selected as the control group.Fluorescence real-time quantitative polymerase chain reaction(RT-PCR) was used to measure the expression of miR-146 a in placental tissues, the online database TargetScan was used to predict the target of miR-146 a and western blot(WB) was used to determine oxidized low-density lipoprotein(OXLDL).The baseline data and blood pressure of the two groups were compared.Results The level of miR-146 a in the placenta tissue of PE group was significantly higher than that of the control group(P<0.05).The database predicted that miR-146 a had a binding site with oxidized low-density lipoprotein receptor 1(OLR1).WB results indicated that the expression of OXLDL protein in the placenta tissue of the PE group was higher than that of the control group, and the quantitative analysis results showed that the difference between the two groups was significant(P<0.05).Conclusion The expression of miR-146 a is elevated in the placenta of PE patients.This may be due to the role of OXLDL receptors in inhibiting OXLDL from blocking nodal signaling.This finding may be a potential therapeutic strategy to block nodal signaling pathways.
