Expression and Significance of GATA3 and Foxp3 mRNA in Peripheral Blood Before and After Radical Resection of Esophageal Cancer

Objective To study the expression and significance of GATA binding protein 3(GATA3)and forkhead box P3(Foxp3)mRNA in peripheral blood before and after radical surgery of esophageal cancer.Methods 196 patients with esophageal cancer were enrolled as case group,while another 60 healthy controls were enrolled as normal group.The expression of GATA3 and Foxp3 mRNA in peripheral blood monouclear cell(PBMC)before and after surgery was detected by quantitative PCR.The expression levels of CD ₃⁺and CD ₄⁺T cells in peripheral blood samples were detected by flow cytometry.The relationship between GATA3,Foxp3 mRNA and clinicopathological characteristics,prognosis of esophageal cancer patients was analyzed.Results The expression levels of GATA3 and Foxp3 mRNA before surgery in case group were higher than those at 1 week after surgery and in normal group,while levels of CD ₃⁺and CD ₄⁺T cells were lower than those at 1 week after surgery and in normal group(P<0.05).There were no significant differences in the levels of GATA3,Foxp3 mRNA,CD ₃⁺ and CD ₄⁺T cells at 1 week after surgery between case group and normal group(P>0.05).The differences in expression level of GATA3 mRNA were statistically significant among esophageal cancer patients with different TNM staging,pathological grading,tumor size and lymph node metastasis(P<0.05).The differences in expression level of Foxp3 mRNA were statistically significant among esophageal cancer patients with different TNM staging and lymph node metastasis(P<0.05).The differences in 1-year survival rate were statistically significant among esophageal cancer patients with different TNM staging,pathological grading,tumor size,lymph node metastasis,GATA3 and Foxp3 mRNA expression(P<0.05).Late TNM staging(RR=1.610)and high-expression(RR=1.473)of Foxp3 were risk factor affecting prognosis of esophageal cancer patients(P<0.05).Conclusion GATA3 and Foxp3 mRNA are highly expressed in esophageal cancer patients,which decrease after radical surgery.Clinically,high-expression Foxp3 should be paid attention to,which is of great significance to the prognosis of esophageal cancer patients.
