摘    要:目的:探讨一氧化氮合酶(eNOS)在白介素10(IL10)和肝再生信息传导中的作用。方法:以70%肝切除制备肝再生模型,注射50%CCl4和脾切除引起肝损伤和机体免疫功能低下,48h后采用逆转录PCR(RTPCR)和点杂交(dotblot)方法检测eNOS和IL10mRNA基因表达,免疫组化LSAB法检测IL10蛋白在肝内的分布,流式细胞仪(FCM)检测再生肝DNA含量,最后采用统计学作再生肝组织DNA含量与IL10和eNOS变化的相关分析,以此判断IL10和eNOS的变化对再生肝的影响。结果:eNOS和IL10对肝再生具有明显的调控作用,无论在生理或在病理状态下都是如此,并且二者呈现一种完全相反的调控方式,同时受到脾脏或全身免疫系统的影响。结论:eNOS的表达同肝细胞DNA的合成呈正相关,而IL10的表达与其呈负相关。

Abstract:Objective:To detect the effects of eNOS and IL 10 on liver regeneration under physiological and pathological condition.Methods:The rat model of liver regeneration was made by partial(70%) hepatectomy method;some rats were injected with CCl 4 and performed splenectomy;the hepatocytes DNA replication were tested by flow cytometry(FCM) method and the gene expression of endothelial cells nitric oxide synthase(eNOS) in liver cells were analysed using RT PCR.The IL 10 gene expression in liver cells was showed by dot blot hybridization method;the synthesize and distribution of IL 10 protein in liver tissue were examined by immune histochemical and morphometric analysis method.Results:eNOS and IL 10 have important immunomodulatory role on liver regeneration both on physiological or pathological conditions,and the effects of eNOS and IL 10 on liver regeneration is different,which play a opposing modulatory effect on liver regeneration,meanwhile,such effect was affected by spleen and general immune system.Conclusions:The expression of eNOS gene promote liver regeneration while the expression of IL 10 gene inhibit it.
