胃癌及癌前病变中端粒酶活性的表达

目的：观察端粒酶活性在胃癌及胃癌前病变细胞和组织中的表达。方法：采用蜡膜介导热启动端粒重复扩增ＰＣＲ模型方法。结果：胃癌细胞株端粒酶活性３株均呈阳性，在３８例胃癌中，３２例端粒酶活性阳性，其中进展期胃癌为３０／３５，早期胃癌为２／３，而邻近胃组织为２／３８，在２９例胃癌前病变中，５例端粒酶活性阳性，其中不典型增生轻度，中度及重度分别为１／８，１／４和１／３，肠上皮化生和胃息肉分别为１／８和１／６，

Telomerase activity in gastric cancer and precancerosis

Aim: To observe the expression of telomeraseactivity in human gastric cancer and precancerous lesions.Methods: Telomerase activity was determined by a wax mediated-hot start telomeric repeat amplification PCR technique (TRAP). Results: Telomerase activity was positivein all 3 gastric cancer cell lines and 32 of 38 gastric cancertissues including 30 of 35 advanced stage and 2 of 3 earlystage cancer tissues, while weak telomerase activity was detected only in 2 of 38 normal gastric tissues. Telomerase activity was expressed in 5 of 29 gastric precancerosis tissues.The order of activities for telomerase in these tissues wassevere (1/3) >moderate displasia (1/4 ) >polyp of stomach(1/6) > mild displasia (1/8 ) = intestinal metaplasia (1/8 ).Conclusion: The expression of telomerase may play a crucialrole in the multistep development of gastric cancer.
