增殖细胞核抗原和甲胎蛋白在暴发性肝功能衰竭动物模型中的表达及意义

目的探讨增殖细胞核抗原(PCNA)和甲胎蛋白(AFP)在暴发性肝功能衰竭(FHF)动物模型中的表达。方法采用硫代乙酰胺(TAA)皮下注射复制大鼠FHF动物模型。取雄性大鼠66只,随机分为正常对照组、模型组。皮下注射TAA 500 mg/kg 2次,间隔24 h。末次注射后分别于3、6、12、24 h各处死8只。鲎试剂法测定血浆内毒素(LPS)含量,判定肠源性内毒素血症情况;赖氏法测定血浆内丙氨酸转氨酸(ALT)含量、苯甲酸钠-咖啡因比色法测定血浆内总胆红素(TB)含量,判定肝功能损伤情况;放射免疫法测定肝匀浆中AFP,判定肝再生过程中的变化;免疫组织化学法检测PCNA在肝细胞中的表达,判定细胞增殖情况。结果血浆中LPS、ALT、TB的水平随着注射TAA时间延长而逐渐升高。肝匀浆中AFP的水平明显升高。免疫组织化学显示:PCNA的表达在FHF发生发展过程中逐渐上调。结论FHF时不仅有肝细胞再生,而且激活了肝干细胞。

Expression and significance of PCNA and AFP in model for fulminant hepatic failure

Objective To investigate the expression of the proliferative cell nuclear antigen(PCNA),alpha fetoprotein(AFP) in model of fulminant hepatic failure(FHF).Methods FHF was induced by subcutaneous injection of TAA.The rats were randomly divided into 2 groups: the group of normal control(N),the group of FHF.FHF rats were not killed till the end of the 3 h,6 h,12 h and 24 h by subcutaneous injection of TAA 500 mg/kg for two days,at the same time.IETM was evaluated by plasma endotoxin;liver function was observed by ALT,TB of plasma;AFP will be demonstrate by immunoradiometrical in the liver tissue;observing hepatic cells regeneration by proliferating cell nuclear antigen(PCNA) immunnohistochemical staining.Results The levels of plasma endotoxin,ALT and TB gradually decreased(P<0.05).The content of AFP in the liver tissue significantly decreased(P<0.05).The immunohistochemical staining showed that the PCNA of liver tissue expression was upregulated in FHF(P<0.05).Conclusion There is not only regeneration of hepatic cells but also activation of hepatic stem cell in the course of FHF occurring.
