环氧化酶-2基因在肝细胞癌中的表达及其意义

摘    要:目的探讨环氧化酶-2(COX-2)在肝细胞癌发生发展中的作用及其意义.方法用免疫组织化学的方法检测41例肝细胞癌组织、16例乙型肝炎后肝硬化组织及5例正常肝脏组织中COX-2蛋白的表达情况,并分析COX-2蛋白表达与肝细胞癌临床生物学特征的关系.结果COX-2蛋白在肝细胞癌和肝硬化组织中的表达率分别为61.0%(25/41)和56.3%(9/16),正常肝脏组织中无表达,与以上各病变组织COX-2蛋白表达差异有显著性(P<0.05);而在肝硬化组织与肝细胞癌组织COX-2蛋白表达差异无显著性(x^2=0.11,P>0.05).COX-2蛋白表达与肝细胞癌组织分化程度、癌肿转移状况有关(x^2=10.3,4.81,P<0.05).结论COX-2蛋白在肝硬化、肝细胞癌组织中表达上调,在肝细胞癌发生发展过程中可能起重要作用.

Abstract:Objective To investigate the expression of cyclooxygenase-2(COX-2) and its significance in human hepatocellular carcinoma.Methods Immunohistochemistry was used to determine the expression of COX-2 protein in human hepatocellular carcinoma(HCC),post-hepatiticcirrhosis tissue(HCL) and normal liver tissues(NL).Results The expression of COX-2 protein was detected in 25 of 41 HCC(61.0%),9 of 16 HCL(56.3%),and none of normal liver tissue.There was significant difference between HCC and NL((P<0.05)),while no significant difference between HCC and HCL((P>0.05)).The expression of COX-2 protein in HCC was correlated with tumor differentiation and metastasis((P<0.05)).Conclusions The expression of COX-2 protein increased in hepatiticcirrhosis and hepatocellular carcinoma.COX-2 protein may play an important role in the development of human hepatocellular carcinoma.
