高纯度獐牙菜苦苷的制备方法

摘    要:目的从川西獐牙菜药材中制备高纯度獐牙菜苦苷。方法川西獐牙菜经醇提水沉后,上清液经HPD-300大孔吸附树脂吸附洗脱后得獐牙菜苦苷粗提物,再用闪式柱色谱对其进行分离。结果制备得到的獐牙菜苦苷,纯度在90%以上,工艺总收率为79.94%。结论本实验建立的制备高纯度的獐牙菜苦苷的方法,可操作性强,重现性较好,工艺收率和产品纯度均较为理想。

Abstract:Objective: To investigate the protective mechanism of Qishenyiqi Pill in acute myocardial ischemia of rats by observing that Qishenyiqi Pill affected the gene expression of gene Ppp3r1 and Atp5d in myocardial ischemia of rats. Methods: Twenty four rats were randomly divided into normal control group, model group, and Qishenyiqi Pill therapeutic group and then the left anterior descending coronary artery (LAD) was ligated to establish the cardiac ischemia model in rats. The respective group differences in Ppp3r1 and Atp5d was detected by real-time fluorescence quantitative PCR. Results: Compared with control group, Ppp3r1 gene expression in model group was increased significantly (P<0.01), while Atp5d decreased (P<0.05).Compared with model group, Ppp3r1 gene expression in Qishenyiqi Pill therapeutic group was decreased significantly (P<0.05), while Atp5d was increased (P<0.01). Conclusion: The protection mechanism of Qishenyiqi Pill in acute myocardial ischemia may be related to the regulation of gene expression in Ppp3r1 and Atp5d..
