白色念珠菌细胞壁不溶性β-葡聚糖抗肿瘤细胞的实验研究

摘    要:目的本研究旨在观察白色念珠菌细胞壁不溶性β-葡聚糖(Candida Albicans Insolubleβ-Glucan,CAIBG)对体外培养的B16恶性黑色素瘤细胞、SGC-7901胃腺癌细胞、SMMC-7721肝癌细胞的生长抑制作用。方法自制CAIBG,采用改良MTT法检测CAIBG在不同时间和不同浓度对体外培养的以上几种肿瘤细胞的生长抑制作用。结果 CAIBG对B16恶性黑色素瘤细胞和SGC-7901胃腺癌细胞有体外生长抑制作用,生长抑制率最高分别为42.8%和59.88%,IC50分别约为1.51mg/μl和0.43mg/μl。CAIBG对SMMC-7721肝癌细胞的体外生长不具有直接抑制作用。结论 CAIBG具有体外直接抑制B16恶性黑色素瘤细胞和SGC-7901胃腺癌细胞生长的作用。CAIBG是具有良好的应用开发前景抗肿瘤药物。

Abstract:Objective In this study,we explored whether CAIBG could suppress the malignant melanoma B16 cells,gastric carcinoma SCG-7902 cells,hepatoma SMMC cells growth in vitro. Methods Insoluble β-glucan was extracted from Candida albicans,and the reforming MTT assay was used to evaluate the growth inhibition effect of different concentrations of CAIBG on malignant melanoma B16 cells,gastric carcinoma SCG7902 cells,hepatoma SMMC cells at different times in vitro. Results CAIBG had inhibitory effects to the malignant melanoma B16 cells,gastric carcinoma SCG-7902 cells in vitro,and the highest growth inhibitory rate of the malignant melanoma and gastric carcinoma were 42. 8% and 59. 88% ,and IC50 were about 1. 5 μg/μl and 1. 81 μg/μl respectively,but had no growth inhibition effect to hepatoma SMMC-7721 cells. Conclusion CAIBG has growth inhibitory effect on malignant melanoma B16 cells and gastric carcinoma SCG-7902 cells. CAIBG is wothy of further research and development.
