双氯芬酸钠滴眼液在家兔房水中的吸收

摘    要: 目的建立房水中双氯芬酸钠的高效液相色谱测定方法,并测定家兔用药后眼房水中药物浓度。方法采用C18色谱柱,流动相为甲醇-水-冰醋酸(80∶20∶0.5),房水用甲醇沉淀蛋白后,上清液挥干,再用甲醇复溶后进样。结果本方法线性范围为10~500 ng·mL-1,最低检测浓度为10 ng·mL-1。3种浓度的平均回收率达到99.7%,日内和日间RSD均小于10%,家兔单剂量应用0.1%双氯芬钠滴眼液50μL后,房水中15 min即可检测到药物,在给药后1h达到最高浓度。结论本方法灵敏,准确,精密。适合于眼房水中双氯芬酸钠的测定。

Abstract:OBJECTIVE To establish a HPLC assay for the determination of diclofenac sodium in aqueous humor following its topical application in rabbits. METHOOS After methanol extraction,the residue was analyzed using a reversed phase C18column with methanol water acetic acid(80∶20∶0.5) as the mobile phase under ultraviolet detection.The aqueous humor samples obtained from rabbits at different interval were mesured. RESULTS The good linearity was achieved over the range of 10~50 ng·mL-1. The mean recovery of three concentrations were 99.7%.The within and between day RSD was less than 10%.Diclofenac sodium was absorbed well through the aqueous humor with the peak concentration at 1 h. CONCLUSION The method was simple, sensitive, accurate and reproducible.It should be applicable for the determination of diclofenac sodium in ocular fluids.
