乌司他丁对百草枯中毒大鼠肺损伤的影响

摘    要:目的观察乌司他丁对百草枯中毒大鼠肺损伤及纤维化的影响,探讨乌司他丁对百草枯中毒大鼠肺的保护机制。方法 SD雄性大鼠90只,体重(180±20)g,随机分为3组:空白对照组(Con组)、百草枯中毒模型组(Mod组)、乌司他丁治疗组(UTI组),每组又再分为5个不同时间点(1 d、3 d、7 d、14 d、21 d,每组6只)。Con组腹腔内注入生理盐水1 ml,其余各组腹腔内注入百草枯溶液18 mg/kg(用生理盐水稀释至1 ml)。0.5 h后,Con组和Mod组:生理盐水1 ml;UTI组:乌司他丁10万U/kg(生理盐水稀释至1 ml),每24 h重复给药一次,直至实验中止。分别于第1天、第3天、第7天、第14天、第21天处死大鼠,采集肺组织,HE染色观察肺组织病理形态学变化,Masson染色观察肺纤维化变化;测定血浆中SOD活性和MDA、HYP含量。结果 HE染色和Masson染色显示:对照组各时间点肺组织呈正常形态结构;Mod组第7天以肺组织炎症为主,第21天出现较明显的肺间质纤维化;UTI组肺组织炎症和肺间质纤维化较Mod组显著减轻。与Con组比较,百草枯染毒各组大鼠血浆SOD活性均显著降低(P<0.05);UTI组血浆SOD活性显著高于Mod组(P<0.05);与Con组比较,百草枯染毒各组血浆MDA和HYP含量均显著升高(P<0.05);UTI组血浆MDA和HYP含量较Mod组显著降低,差异有统计学意义(P<0.05)。结论乌司他丁能通过减轻炎性细胞与氧自由基对肺的损伤作用而发挥对肺的保护作用。

Abstract:Objective To observe the effect of UTI to paraquat intoxication rats lung injury and fibrosis influence, and investigate the mechanism of the effect of UTI. Methods The male rats weighting (180&#177;20)g were randomly divided into three groups, there were 30 rats in every group:control group(Con group), model group(Mod group), UTI group(UTI group), 1 d, 3 d, 7 d, 14 d and 21 d each again divided into five subgroups(n=6). 1 ml saline was injected intraperitoneally into rats of control group, as to the other groups, paraquat 18 mg/kg (diluted with saline to 1 ml) was injected intraperitoneally. Half an hour later, according to the following dose:Con group and Mod groups:normal saline 1 ml;UTI Group:UTI 10 kU/kg; The intervention drugs were diluted to 1 ml with saline, all by the way of intraperitoneal injection, repeated administration of once every 24 h, until the end of the experiment. On the 1st, 3rd, 7th, 14th and 21th day after administration, animals were killed to collect the lung tissue, plasma to detect the pathological changes of lung tissue, detect the activity of SOD and the content of MDA and HYP in plasma. Results HE staining and Masson staining: the morphology of lung tissue was normal in Con group at different time points;in Mod Group, there was mainly lung inflammation on 7th day, while on 21th day, there was obvious pulmonary interstitial fibrosis; the lung inflammation and pulmonary fibrosis of UTI group significantly reduced compared with Mod group;Compared with Con group, the activity of SOD in&amp;nbsp;plasma of rats exposured to paraquat was significantly decreased (P〈0.05); but the activity of SOD in plasma UTI group was significantly higher than that of Mod group (P〈0.05); Compared with the Con group, the content of MDA and HYP in plasma exposured to paraquat was significantly increased (P〈0.05). The content of MDA and HYP in plasma of UTI group was significantly lower than that of Mod group, the difference had statistical significance
