IL-27对慢性丙肝患者外周血Th1／Th17水平调节作用的研究

目的探讨IL-27体外对慢性丙肝患者外周血Th1／Th17水平调节作用。方法分离慢性丙型肝炎（ChronicHepatitisC，CHC）患者和健康捐献者（HealthDonor，HD）外周血单个核细胞（Pefipheral DloodMononuclearCells，PBMC），将IL-27加入CHC患者及HD的PBMC并培养72h，CHC患者和HD外周PBMC组不做为空白对照，应用流式细胞仪检测IL-27刺激前后不同组PBMC中CD4＋T细胞内IFN．1和IL-17的变化情况，计算Th1、Th17细胞比例，观察体外IL-27对CHC患者和HD外周血Th1／Th17的影响。结果IL-27刺激48h后Th1细胞亚群变化不明显，Th17细胞比例明显下降（P=0．01）。72h后CHC＋IL-27组Th1细胞比例较CHC组升高（P=0．04）．Th17细胞比例仍表现为下降趋势（P=0．02），变化较HD组更加明显。结论IL-27可以刺激慢性丙肝患者Th1／Th17的变化，表现为IL-27提高患者免疫系统中Th1细胞的比例，降低Th17细胞比例。

Effect of IL-27 on Level of Th1/Th17 in Peripheral Blood of Patients with Chronic Hepatitis C

Abstract： Objective To evaluate the effect of IL-27 on the level of Th1/Th17 in patients with chronic hepatitis C （CHC） in vitro. Methods The peripheral blood mononuclear cells （Peripheral Dlood Mononuclear Cells, PBMC ） of patients with CHC and health donor（HD） were isolated. IL-27 was added into PBMC and cultured it in vitro with PBMC alone as control. Flow cytometry was used to detect the IFN-γ, and IL-17 changes in CD4＋ T cell of PBMC in each group. Then the ratio of Th1, Th2 cells were calculated, and the effect of IL-27 on Th1/Th17 in patients CHC in vitro was assessed.Results By stimulating for 48 h, IL-27 could inhibit proliferation of lymphocyte in vitro. Compared to baseline, C HC ＋IL-27 group did not showed a rising trend in Th 1 cell, while Th17 cells present down trend （P = 0.01 ）. After 72 h, CHC＋IL-27 group showed a rising trend in Th1 cell （P= 0.04） and Th17 cells still present down trend （P= 0.02） compared with that of the control group and HD group. Conclusion IL-27 played a regulatory role in Th1/Th17 in patients with CHC in vitro, by rising the ratio of Th1 cells and decreasing the Th17 cells.
