氟伐他汀对骨质疏松大鼠骨折愈合的作用研究

摘    要:目的探讨氟伐他汀对卵巢切除大鼠股骨干骨折愈合的作用。方法 6个月龄雌性SD大鼠50只,随机分为5组,每组10只,分为假手术组、双侧卵巢切除术组、假手术+骨折+生理盐水组、卵巢切除+骨折+生理盐水组、卵巢切除+骨折+氟伐他汀组。骨折组的大鼠均采用右股骨中断横形骨折,髓内针固定,氟伐他汀给药组采用大鼠灌胃10mg/(kg.d)。实施药物灌胃3个月后,处死大鼠,取样分析血清生化指标、骨密度以及骨形态,CR摄片观察各组骨折愈合情况,放射免疫分析法测血清BMP-2含量。结果与假手术组相比,卵巢切除使大鼠血清钙和碱性磷酸酶的含量降低,并且降低了骨密度,使骨形态恶化。两组间BMP-2表达量差异无统计学意义(P〉0.05);与生理盐水灌胃组相比,氟伐他汀可以增加大鼠血清钙和碱性磷酸酶的含量,阻止骨密度的降低,并显著提高血清BMP-2表达水平。结论骨质疏松降低大鼠血清钙和碱性磷酸酶含量,降低骨折愈合质量,骨质疏松对骨折愈合过程的影响与BMP-2的表达无明显相关性;氟伐他汀对骨质疏松大鼠具有明显的促骨折愈合作用,而且与BMP-2的表达有显著相关性。

Abstract:Objective To evaluate the effects of fluvastatin on fermoral fracture healing in bilaterally ovariectomized rats. Methods Fifty six-month-old Sprague-Dawley rats were divided randomly into 5 groups,with 10 in each group.They were shamed operaded group,bilaterally ovariectomized group, shamed operaded and fermoral fracture and administration of N.S group, bilaterally ovariectomized and fermoral fracture and administration of NS group, bilaterally ovariectomized and fermoral fracture and administration of fluvastatin group. Fluvastatin was administrated for 3 months 10 mg/(kg·d).All rats were sacrificed,analysis the content of serum calcium and alkaline phosphatase, the expression of BMP-2 was measured by using Rat BMP-2 ELISA kit;The fracture healing in each group was observed by CR radiography.Results Compared with sham operation group, the content of serum calcium and alkaline phosphatase were reduced because ovariectomy,and and reduced bone mineral density, deterioration of the bone histomorphometry.BMP-2 expression between the two groups showed no statistical difference (P〉 0.05);Compared with the saline group, fluvastatin can increase serum calcium and alkaline phosphatase levels, preventing bone mineral density decreasing, and significantly increase the serum levels of BMP-2 expression. Conclusion Osteoporosis lower serum calcium and alkaline phosphatase, reduces the quality of fracture healing; The influencing effects of osteoporosis to fracture healing had no significant correlation BMP-2 expression, Fluvastatin has obvious promoting fracture healing effects on osteoporosis rats, but also the expression of BMP-2 was significantly correlated.
