高脂高糖大鼠模型建立及血清PAI-1、APN表达的研究

摘    要:目的:探讨高脂高糖大鼠模型的建立方法,并观察血清PAI-1、APN的表达。方法:利用高糖高脂饲料及给予L-NAME建立高脂高糖模型大鼠,观察模型大鼠血清葡萄糖(GLU)、总胆固醇(TC)、三酰甘油(TG)、高密度脂蛋白(HDL-C)、低密度脂蛋白(LDL-C)、纤溶酶原激活物抑制物-1(PAI-1)、脂联素(APN)的表达。结果:造模72 h后测量空腹血糖≥7.8 mmol·L-1,模型组血清GLU、TC、TG、LDL-C含量均较正常对照组明显升高(P〈0.01),提示造模成功。高脂高糖模型大鼠血清PAI-1明显升高、APN明显下降,较正常对照组有显著性差异(P〈0.01)。结论:利用高脂高糖饮食和灌胃给予LNAME,加上链脲佐菌素(STZ)诱导,成功制作了高脂高糖症大鼠模型。高脂高糖模型大鼠血清PAI-1水平明显上调、APN水平明显下调。

Abstract:Objective:To explore the rat model of high fat and sugar to create and serum PAI-1, APN expression. Methods:High sugar, high fat diet and given L-NAME rat model of high fat and sugar, to observe the expression of the rat model of serum GLU and TC, TG, LDL-C, HDL-C content and serum of PAI-1, APN. Results : Modeling measured after 72h fasting plasma glucose≥ 7.8mmol / L, serum GLU and the TC,TG and LDL-C content of the model group compared with the normal control group was significantly increased (P 〈 0. 01 ), suggesting that the model was successful. Serum of PAI-1 was significantly increased APN decreased significantly, compared with control group significant difference( P 〈 0. 01 ). Conclusion:Under the experimental conditions, the use of high-fat high-sugar diet and oral L-NAME, coupled with streptozotocin(STZ)-induced, and successfully produced a high-fat high-sugar disease rat model. The high fat and sugar model of serum PAI-1 levels were significantly raised ,the APN levels were significantly reduced.
