摘    要:松潘乌头总碱(TAS)ip,ig,sc给药均呈剂量依赖性抑制小鼠热板、扭体及嘶叫反应。TAS的镇痛作用明显强于高乌甲素。连续给药7 d,TAS的镇痛作用无耐受现象。小鼠跳跃试验和竖尾试验均为阴性结果,提示TAS是一种不同于吗啡的非成瘾性镇痛剂。

Abstract:Aconitum Sungpanense hand-mazz is a traditional Chinese herb. In this study, the total alkaloids of aconitum Sungpanense hand-mazz (TAS) was utilized to in- vestigate the postulated effect. The LD_(50) of ip TAS was 1. 84 ± s 0. 28 mg .kg~(-1) and of ig TAS was 8.66±s 0.58 mg . kg~(-1) in mice. Analgesic effect of TAS was demonstrated in mice by using the analgesic assays of writhing, hot plate and squeak. The experimental re- sults showed that the effect of writhing, hot plate and squeak reaction of mice was markedly suppressed by TAS at the dosage of ip or sc 0.25 -0.60 mg .kg~(-1) and ig 0. 50-3. 00 mg . kg~(-1). The peak time was 0.5h after administration of ip TAS and 3. 5 h after ad- ministration of ig TAS. The duration of analgesic effect of ip TAS was 1.5 h and of ig TAS was 3.0 h. The relative analgesic effect of ip TAS was found to be 13-15 times as potent as ip lappaconitine. No tolerance of analgesic effect was found after daily ip of TAS 0.62 mg .kg~(-1) for 7 d in mice assayed with hot plate method. In mice, ip TAS 0.25 or 0.60 mg.kg~(-1) exhibited a sedative effect, while ip morphine 15 or 20 mg .kg~(-1) caused a significant Straub' s tail re- action and motor activity. In nalorphine - challenge test, no jumping response was ob- served in mice treated with TAS 4,9 or 12.0 mg. kg~(-1). The results indicated that TAS induced neither morphine -like tolerance nor physical dependence.
