NOS抑制剂对局灶性缺血再灌流大鼠脑神经细胞凋亡的影响研究

目的 研究一氧化氮合酶（ＮＯＳ）抑制剂在局灶性脑缺血再灌流过程中对神经细胞的作用机理,特别是ＮＯＳ抑制剂（ＮＮＬＡ）与细胞凋亡的关系。方法 利用末端标记法,测定ＮＮＬＡ干预后局灶性脑缺血鼠脑组织中神经细胞的凋亡情况。结果 大、小剂量的ＮＯＳ抑制剂分别具有增加或减少阳性凋亡细胞出现率的作用。结论 给予不同剂量的ＮＯＳ抑制剂与单纯缺血再灌流相比,对脑神经细胞的凋亡出现率有不同的影响。

Effect of inhibitor of nitric oxide synthase on neuronal apoptosis in the focal cerebral ischemic reperfusion in rats

Objective To study the mechanism of neurocyte on inhibitor of nitric oxide synthase (NOS) in the focal cerebral ischemic reperfusion and the relationship between inhibitor of NOS and apoptosis especially. Methods Using terminal deoxyribo-nucleotide transferase-mediated dUTP-biotin nick end labeling(TUNEL) method,the changes of apoptosis cells in the rat's brain tissue on the focal cerebral ischemic reperfusion were determined after NNLA intervention. Results Big dose of inhibitor of NOS could increase positive neuronal apoptosis. Small dose of inhibitor of NOS could decrease positive neuronal apoptosis. Conclusion NOS inhibitors of different doses were administered as compared with cerebral ischemic reperfusion,there was different effect in the prevalence of brain neuronal apoptosis.
