人脑胶质瘤中Ets-1表达与肿瘤血管生成、癌细胞增殖浸润的关系

摘    要:目的:研究胶质瘤组织中原癌基因蛋白Ets-1、微血管密度(MVD)和细胞增殖核抗原(k-i 67)的表达及其关系,探讨Ets-1在胶质瘤发生、发展、侵袭转移中的作用及临床意义。方法:采用免疫组化双染法和SP法检测Ets-1表达及MVD和K-i67在63例胶质瘤(实验组)和10例正常脑组织(对照组)中的表达,并进行相关性分析。结果:实验组中Ets-1表达阳性率为38.1%,高于对照组(χ2=5.204,P<0.05),Ⅲ~Ⅳ级组织中的Ets-1表达随临床分级(R=0.59,P<0.001)的增高而呈增高趋势。实验组MVD计数(33.7±20.4)明显高于对照组(5.6±3.9)(F=18.31,P<0.001),MVD值随Ets-1表达的增强而增高(P<0.001)。实验组中K-i67表达率为61.9%,与对照组比有统计学意义(χ2=13.29,P<0.001)。K-i67表达随Ets-1表达增强而增高(R=0.46,P<0.001)。结论:Ets-1、MVD、k-i67可作为胶质瘤恶性程度,浸润转移等生物学行为的重要参考指标。

Abstract:Objective:To investigate the expressions of proto-oneogene protein(Ets-1),microvessel density(MVD),cell proliferation nuclear antigen(Ki-67)and their relationship in glioma tissue,and to explore the effect of Ets-1 in the development,growth and invasion in glioma.Mothods:Double-labeling immunohistochemistry method and immunohistochemical staining-SP method were used to detect the expression of Ets-1,MVD,Ki-67 in 63 cases with glioma(experiment group)and 10 cases of normal brain tissue(control group).Results:The positive rates of Ets-1 was 38.1% in the experiment group.The expression of Ets-1 was increasing with the clinical grade rising.MVD was 33.7±20.4 in the experiment group and 5.6±3.9 in the control group(F = 18.31,P0.001).MVD was increasing with the expression of Ets-1 aggrandizing.Ki-67 was highly expressed in the experiment group(61.9%)than control group(χ2 = 13.29,P0.001).There was significant correlation between the expression of ki-67 and increased expression of Ets-1(R = 0.46,P 0.001).Conclusion:Ets-1,MVD,Ki-67 may be a valuable marker to evaluate the cancer development,growth and invasion in glioma.
