结直肠癌组织中GINS复合物的表达及其临床意义

摘    要:目的探讨GINS复合物在结直肠癌组织中的表达及其临床意义。方法运用实时荧光定量PCR方法检测76例结直肠癌标本中GINS复合物4个基因PSF1、PS砣、PSF3和SLD5的mRNA表达情况.并分析其与结直肠癌患者临床病理特征及预后的关系。结果结直肠癌组织中PSF1、PSF2、PSF3和SLD5mRNA的相对表达量(0.001853±0.000651、0.007757±0.004260、0.000967±0.000481、0.003248±0.001721)均明显高于正常黏膜组织(0.000352±0.000169、0.002951±0.001216、0.000472±0.000271和0.001675±0.001156)(均P〈0.01)。PSF1mRNA表达与肿瘤大小(p〈0.01)有关;PSF2mRNA表达与患者年龄(P〈O.05)和淋巴结转移(P〈0.05)有关;PSF3mRNA表达与患者临床病理特征无关(P〉0.05);SLD5mRNA表达与患者淋巴结转移(P〈0.01)有关。PSFl、PSF2和SLD5高表达组患者的5年生存率分别为57.1%、54.3%和54.3%,均明显低于低表达组患者的77.1%、80.O%和80.0%,差异均有统计学意义(均P〈0.05)。Cox回归分析结果提示,PSFlmRNA表达是影响结直肠癌患者预后的独立因素之一(P〈0.05)。结论GINS复合物在结直肠癌组织中高表达,且与患者临床病理特征有密切关系.其中PSFlmRNA表达可作为预测结直肠癌患者预后的生物学指标。

Abstract:Objective To investigate the expression and clinical significance of GINS complex in colorectal cancer (CRC). Methods The expression level of GINS complex including PSF1, PSF2, PSF3 and SLD5 in CRC specimens (n=76) were detected by real-time fluorescent quantitative polymerase chain reaction. The association of GINS complex with clinicopathological parameters and prognosis of CRC patients were analyzed.Results The relative expression level of PSF1, PSF2, PSF3, and SLD5 mRNA in CRC tissues was 0.001 853±.0000 651, 0.007 757±.0004 260, 0.000 967±0.000 481 and 0.003 248 ±.0001 721, which was significantly higher than that in normal colorectal mucosa tissues (0.000 352±.0000 169, 0.002 951±.0001 216, 0.000 472±.0000 271, and 0.001 675±0.001 156) (all P<0.01).PSF1 mRNA expression was associated with tumor size (P<0.01), and PSF2 mRNA expression with age (P<0.05) and lymph node metastasis (P<0.05). No correlations between PSF3 mRNA expression and clinicopathological parameters were observed. SLD5 mRNA expression was associated with lymph node metastasis (P<0.01). Patients with high expression of PSF1, PSF2 and SLD5 had worse 5-year overall survival rate (57.1%, 54.3%, and 54.3%) than those with low expression (77.1%, 80.0%, and 80.0%)(all P<0.05). Multivariable Cox regression analysis indicated that PSF1 mRNA expression (P<0.05) was an independent factor associated with prognosis of colorectal cancer. Conclusions Overexpression of GINS complex in CRC is associated with clinicopathological characteristics and prognosis of colorectal cancer. PSF1 expression is prognostic for CBC patients.
