摘    要: 目的 建立人血浆中利奈唑胺的HPLC测定方法方法 采用氯霉素为内标,用5%三氯乙酸-甲醇溶液(1∶1)沉淀蛋白,色谱柱为Symmetry C18 (4.6 mm×150 mm,5 μm),流动相为乙腈-水 (28∶72),流速为1.0 mL·min-1,柱温30 ℃,检测波长254 nm。结果 利奈唑胺在0.25~50 mg·L-1内线性关系良好,r=0.999 7,最低检测浓度为0.05 mg·L-1,平均绝对回收率95.68%,方法回收率在100%~102%之间。高、中、低3种浓度的日内、日间RSD均小于6%。结论方法简单、快捷、灵敏、准确,适用于利奈唑胺临床血药浓度的监测。

Abstract:OBJECTIVE To establish a HPLC method for the determination of linezolid in human plasma. METHODS The analytes in plasma were extracted by protein precipitation using 5% trichloroacetic acid and methanol(1∶1), and chloramphenicol was used as the internal standard. The separation was performed on a Symmetry C18 Column (4.6 mm×150 mm, 5 μm) at 30 ℃. The mobile phase consisted of acetonitrile-water (28∶72) with a flow rate of 1.0 mL·min-1-1.The UV detection wavelength was 254 nm. RESULTS The calibration curves of linezolid showed good linear regression in the range of 0.25-50 mg·L 1 (r= 0.999 7). The limits of detection was 0.05 mg·L-1. The mean absolute recovery was 95.68%, and the method recovery was 100%-102%. Intra-and inter-day variations were less than 6%. CONCLUSION The established method is simple,sensitive and accurate for determining linezolid level in human plasma.
