药根碱诱导白血病K562细胞凋亡的研究

摘    要: 目的 研究药根碱对白血病K562细胞的生长抑制及凋亡作用,探讨药根碱诱导K562细胞凋亡的可能途径。方法 采用四甲基偶氮唑盐(MTT)法检测药根碱对K562细胞的生长抑制作用;吖啶橙(AO)/溴化乙锭(EB)双染法、碘化丙啶(PI)染色法观察药根碱对K562细胞凋亡的诱导作用;利用活细胞高内涵成像系统检测细胞内线粒体膜电位以及氧自由基的变化。结果 药根碱可明显抑制K562细胞的生长(IC50 = 90 μmol·L-1 )。AO/EB染色可明显看到凋亡细胞和死亡细胞。PI染色显示,药根碱使K562细胞周期阻滞在G0/G1期。活细胞高内涵成像系统检测结果显示,37、75、150 μmol·L-1 药根碱作用后,细胞内活性氧荧光强度由1 305.1±15.3上升为26 243.6±165.3、30 106.7±153.2、34 682.6±174.1,线粒体膜电位由1 025.3±20.1下降为484.6±10.2、202.9±6.3、153.1±5.3。结论 药根碱能明显抑制白血病K562细胞生长,促进K562细胞凋亡,且其可通过增加细胞内活性氧、降低线粒体膜电位这一线粒体信号传导通路来诱导K562细胞凋亡。

Abstract:OBJECTIVE To study the effect of jatrorrhizine on the proliferation and apoptosis of K562 cells and analyze the pathway in which jatrorrhizine induces apoptosis of K562 cells. METHODS The inhibitory effect of jatrorrhizine on the proliferation of K562 cells was determined by MTT assay.The effects of jatrorrhizine on the apoptosis of K562 cells were studied by AO/EB and PI staining. The changes of mitochondrial membrane potential and intracellular reactive oxygen were detected with live cell high-content imaging systems. RESULTS The proliferation of K562 cells was inhibited in dose-dependent manner. The changes in K562 cells morphology were clearly obtained by AO/EB staining. PI staining assay indicated that jatrorrhizine arrested K562 cells in the Go/Gl phase.By live cell high-content imaging systems, jatrorrhizine increased intracellular reactive oxygen obviously and decreased mitochondrial membrane potential. CONCLUSION Jatrorrhizine could inhibit the growth of leukemia K562 cells and induce the apoptosis of those in a mitochondrion-dependent manner .
