摘    要:目的观察缺氧预适应(PC)对体外培养的心肌细胞的保护作用以及不同时相的PC对心肌细胞摄取201T1的影响。材料和方法在原代培养的乳鼠心肌细胞基础上,建立早期缺氧预适应(EP)和晚期缺氧预适应(DP)模型,观察EP和DP条件下心肌细胞存活和摄取210T1情况。结果与缺氧/复氧组相比较,EP组细胞存活率提高20%,DP组提高12%,均有显著差异(P<0.01)。EP组心肌细胞在复氧1小时后对210T1的摄取接近正常组(P>0.05),DP组心肌细胞在复氧1小时后210T1的摄取量低于正常组(P<0.05),与缺氧/复氧组相比无显著性差异(P>0.05)。结论EP和DP能够保护体外培养乳鼠心肌细胞,两时相摄取210T1表现不同,可能与其不同的保护机制有关。

Abstract:Purpose: To study the protection on cultured cardiomyocytes of hypoxic preconditioning (HPC) and to investigate the effects at different time of HPC on Thallium-201 uptake in vitro. Materials and Methotls: In the early protection (EP) and delayed protection (DP) models, the effects of HPC on the viabilty of primary cultured neonatal rat cardiomyocytes and thallium-201 uptake were observed. Results: Compared with normal contral group, the viable myocytes in EP group increased 20 % and in DP increased 12% (P<0.01). The uptake of thallium-201 in group of EP reached normal level (P>0.05) at for after reoxygenation, while that in group of DP kept low (P <0.05) in comparison with normal contral group and similar to the uptake at the group of hypoxia/reoxygenation (P>0.05). Conclu non: EP and DP of HPC existed in primary cultured neonatal rat cardiomyocytes and the effects at the two phases on the uptake of thallium-201 were different, which may be related to their different protective mechanisms.
