摘    要:目的 评价以川贝母为主要组分的两种保健食品配方对大鼠慢性咽炎模型的清咽功能,并探讨其潜在作用机制。方法 85只SPF级雄性SD大鼠(4~5周龄),随机分为阴性对照组(12只)和造模组(73只)。通过咽部喷注2.5%氨水和滴注松节油建立慢性咽炎模型,以咽部组织炎性病理评分≥2分作为评判标准。将造模成功的大鼠随机分为阴性对照组、模型组、阳性对照组(人体推荐剂量5倍的西洋参川贝枇杷膏)、两种配方的低和高剂量组(人体推荐剂量的5倍和20倍),灌胃给予。配方1含川贝母、麦冬、金银花和余甘子,配方2含川贝母、麦冬、青果和陈皮。给药30 d后,取心肝脾肾称重,全血检测白细胞、中性粒细胞和淋巴细胞数,咽部组织HE染色,血清和咽部组织检测炎症因子水平。结果模型组咽部组织炎性病理评分高于阴性对照组,且评分≥2分,造模成功。阳性对照组和配方2高剂量组咽部组织病理评分低于模型组,阳性对照组血清中TNF-α水平低于模型组,配方2高剂量组血清中IL-1β和TNF-α水平低于模型组。结论 咽部喷注2.5%氨水和滴注松节油可建立大鼠慢性咽炎模型。阳性对照组和配方2显著改善了大鼠咽部炎性损伤,其抗炎机制与血清中IL-1...

Abstract:Objective To evaluate the throat-clearing function of two functional health food with Fritillaria cirrhosa D.Don as the main component in a rat model of chronic pharyngitis and explore its potential mechanism. Methods In total 85 SPF male SD rats of 4 to 5 weeks old were randomly divided into negative control group (n=12) and model group (n=73). The model of chronic pharyngitis was established by injecting 2.5% ammonia water and turpentine into the pharynx, and the inflammatory pathological score of pharynx tissue ≥ 2 was used as the evaluation standard. The rats with successful modeling were randomly divided into negative control group, model group, positive control group (5 times recommended dose of American ginseng Chuanbei loquat ointment), and low and high dose groups (5 times and 20 times of recommended dose). Formula 1 contained Fritillaria cirrhosa D.Don, Ophiopogon japonicus (L. f.) Ker-Gawl, Lonicera japonica Thunb, and Phyllanthus emblica L., and Formula 2 contained Fritillaria cirrhosa D.Don, Ophiopogon japonicus (L. f.) Ker-Gawl, Canarium album Raeusch, and Citrus reticulata Blanco. After 30 days of administration, the heart, liver, spleen, and kidney were weighed, and the number of leukocytes, neutrophils and lymphocytes in whole blood were detected. HE staining was performed in pharyngeal tissue, and the level of inflammatory factors in serum and pharyngeal tissue were detected. Results The inflammatory pathological score of pharyngeal tissue in the model group was higher than that in the negative control group, and the score was ≥ 2. The histopathological score of pharynx in the positive control group and formula 2 high dose group was lower than that in the model group, the serum TNF- α level in the positive control group was lower than that in the model group, and the serum IL-1 β and TNF- α levels in the high dose Formula 2 group were lower than those in the model group. Conclusion Pharyngeal injection of 2.5% ammonia and turpentine can establish the model of chronic pharyngitis in rats. The positive control group and Formula 2 significantly improved the inflammatory injury of pharynx in rats, and the anti-inflammatory mechanism is related to the decrease of serum levels of IL-1 β and TNF- α.
