| 摘 要： | 目的了解溶血磷脂酸（LPA）对胃癌腹膜间皮细胞（PMC）分泌血管内皮生长因子（VEGF）的调控关系。方法体外分离培养PMC并接受LPA的诱导，ELISA检测VEGF的分泌量，随后用LPA的两种拮抗剂PTX和Ki16425分别孵育，观察VEGF水平的变化。结果PMC在受到LPA的刺激，产生的VEGF呈剂量依赖性增长，经PTX或Ki16425的拮抗，LPA诱导PMC生成VEGF的量明显下降。结论LPA诱导VEGF的分泌在胃癌腹腔扩散中起着重要的作用。

| Abstract: | Objective To examine the relation between lysophosphatidie aeid（LPA） and vascular en- dothelial growth faetor（VEGF） in cultured peritoneal mesothelial eells（PMC） in gastric eaneer. Methods Human PMC stimulated with LPA were isolated and cultured in vitro. The seeretion changes of VEGF in PMC were examined by ELISA. Then PMC was incubated with PTX or Ki16425, which were antagonists of LPA. Effect of PTX on LPA-indueed VEGF secretion was determined. Results VEGF secretion in LPA-indueed PMC increased in a dose-dependent manner. After PMC were treated with PTX or Ki16425, VEGF secretion were significantly reduced. Conelusion LPA-indueed VEGF secretion may have an important role in the growth of cancer cells disseminated in the peritoneal cavity. |
