The relation between polyunsaturated fatty acid and adipocytokines on NASH rats

Abstract:Objective To observe the efficacy of polyunsaturated fatty acid(PUFA) on adipocytokines in rats with NASH. Methods Forty-four rats were randomly divided into 4 groups (control/ NASH/ lower therapy/ higher therapy group). Rats in control group were fed with normal diet,and the other three with high-fat diet. co-3 unsaturated fatty acid was added to therapy group from 8th week to 12th week(0.5 g/d and 1.0 g/d). The body/liver weight, HDL, LDL, TC and TG in liver of rats in each group were measured. Adipocytokines were detected by ELISA and real time fiourescence quantitive PCR. Results NASH model was established successfully. The weight, HDL, LDL, TC and TG were decreased in lower/ higher therapy group, as compared with those in rats with NASH. Changes of HDL, LDL and TC showed significant difference in higher therapy group(P〈0.05, P〈0.01, P〈0.01 ). Lower/higher therapy group showed lower leptin, resistin and higher adiponectin levels than those in NASH group. Changes depended on dosage of PUFA. Adipocytokines in liver differed between higher group and lower one(P〈0. 05), among which resistin and adiponectin mRNA expression was of statistical significance (P〈 0. 01 ). Conclusion PUFA could improve lipid metabolism and regulate the expression of adipocytokines on NASH rats.
