正交试验优选罗勒子中总黄酮的提取工艺

摘    要:目的优选罗勒子中总黄酮的提取工艺。方法以芦丁为标准品,通过紫外分光光度法测定罗勒子中总黄酮含量,通过单因素试验和正交试验考察不同因素和水平对提取工艺的影响。结果甲醇浓度对罗勒子中总黄酮含量影响最大,其他三个因素对样品含量的影响大小依次是:料液比>提取次数>提取时间。结论最佳提取工艺条件为甲醇浓度80%,固液比1∶10,提取1 h,提取1次。

Abstract:Objective To optimize the extract technology of the total favonoids from the fruit of ocimum basilicum. Methods Determination of total favonoids in fruit of ocimum basilicum by UV, the content of favonoids was computed with rutin as standard substance, and the influence of different factors and levels on the extract technology were investigated by means of simple factor experiment and orthogonal experiment. Results Methanol concentration had great impact on the target. The order of the other 3 factors that affected the fiavonoid extractions was amount of ethanol〉number of extractions〉extraction time. Conclusion The optimum extraction technology was as follows: 80% Methanol concentration 10 times the weight of the dried material, reflux extracting total favonoids once, 1 h at each time.
