半边莲生物碱对内皮素诱导损伤的人血管内皮细胞纤溶系统的影响

摘    要:目的:研究半边莲生物碱对内皮素诱导损伤的人血管内皮细胞的保护作用。方法:常规提取半边莲生物碱,培养人血管内皮细胞.ELISA法测定细胞培养上清中组织纤溶酶原激活物(t-PA)和纤溶酶原激活物抑制物-1(PAI-1)的含量。结果:内皮素组PAI-1含量显著高于正常对照组(P〈0.01).t-PA含量在数值上低于正常对照组,但无统计学意义。半边莲生物碱各组PAI-1含量均低于内皮素组(P〈0.01).与内皮素B受体拮抗剂BQ7S8作用相似。半边莲生物碱低剂量组t-PA含量高于内皮素组(P〈0.01).高、中剂量组t-PA含量虽然有增高趋势.但无统计学意义。结论:内皮素能够使人血管内皮细胞释放PAI-1增加.半边莲生物碱通过抑制该效应而起到保护血管内皮细胞的作用。

Abstract:Objective: To observe the injury effect of endothelin on human vascular endothelial cells(HVECs) and study the inhibitory role of Lobelia Chinensis Lour alkaloids (LCLAs) on this effect. Methods: Alkaloids were extracted from Lobelia Chinensis Lour. The concentrations of tissue-type plasminogen activator (t-PA) and plasminogen activator inhibitor-1 (PAI-1) in supernatants were assayed using the enzyme linked immunosorbent assay technique. Results: The concentrations of PAI-1 were higher in endothelin groups than in control group (P<0.01). LCLAs lessened the release of PAI-1 significantly from HVECs injured by endothelin (P<0.01). Their effects were similar to those of endothelin B receptor antagonist,BQ788. Conclusion: LCLAs can protect HVECs against the injury of endothelin.
