| 摘 要： | 目的检测马齿苋黄酮提取液中染料木苷的含量。方法用反相高效液相色谱（RP-HPLC）检测马齿苋黄酮部位中染料木苷的含量。色谱柱为SunfireC18column（250.0mm×4.0mm，5.0μm），流动相为甲醇：冰醋酸：水＝45：1：55，流速为0.8ml/min，柱温25℃，柱压2144psi，检测波长为259nm。结果马齿苋黄酮部位中染料木苷与标准品中染料木苷保留时间一致，染料木苷进样量在0.0625～2.0000ng范围内线性关系良好（r＝0.9999），平均回收率98.30％，RSD＝0.57％（n＝5）。结论RP-HPLC测定方法简便，结果可靠，含生药6kg/L的马齿苋黄酮提取液中染料木苷含量为1.66mg/L。

| Abstract: | Objective To develop reversed phase high-performance liquid chromatography （RP-HPLC）method for the determination of genistin in flavone extract from Portulaca oleracea L. .Methods The chromatographic separation was performed on a Sunfire C18 column（250.0 mm × 4.0 mm ,5.0μm）and eluted with a mobile phase of MeOH： HAC： H2 O（45：1：55）.The detection wavelength was set at 259 nm and column temperature was set at 25 ℃.The flow rate was 0.8 ml/min.Results Stable linear relation between the peak area and injected amount ex-isted when the amount was within 0.062 5 -2.000 0 ng for genistin （r= 0.999 9）.The average recovery was 98.30% and the RSD was 0.57% （n=5）.Conclusion The HPLC method was accurate and sensitive for the deter-mination of genistin in flavone extract from Portulaca oleracea L. . |
