含氧化苦参碱血清的制备与抗肿瘤作用关系研究

摘    要:目的研究不同给药方式,不同采血时间获得的含氧化苦参碱血清对卵巢癌细胞H08910的生长抑制作用,探讨含药血清的制备方法。方法选择SD大鼠,采用肌注、灌胃、腹腔注射等形式给予氧化苦参碱药物,分别在不同时间取血,通过TLC法判断血清的化学成分,MTT法确定这些含药血清对卵巢癌细胞H08910的生长抑制作用。荧光显微镜观察细胞的形态学的改变。结果不同的给药方式获得的含药血清均含有氧化苦参碱,每天肌注给药两次,连续给药3d,末次给药后1h采血获得的含药血清对卵巢癌细胞的抑制作用最强,荧光显微镜下观察到典型的细胞凋亡形态学变化。结论经代谢后含氧化苦参碱血清仍具有抑制肿瘤细胞生长的作用。其抑制作用与给药形式、采血时间有明显的关系。

Abstract:Objective] To investigate the inhibition effect of Oxymatrine serum on human ovarian cancer HO8910 cells at various blood-collection times and different route of drug administration. The method of preparing drug-containing serum was also explored. Method] The serum of SD rats were collected at different time after im, ig and po administration of Oxymatrine, separately. The chemical components of the serum was detected by thin layer chromatogram (TLC). The effect of the drug-serum on the proliferation of culture human ovarian cancer HO8910 cells was analyzed with MTT assay. Morphological changes of the cells were observed under fluorescent microscope. Results] The drug-serum still contains Oxymatrine after Oxymatrine was biotransformated in body. The drug containing serum which was taken 1 h after im administration of Oxymatrine at last time for 3 consecutive days, two times every day has strong inhibition effect on human ovarian cancer HO8910 cells. Conclusion] The serum after Oxymatrine was biotransformated in body and still have the effects of inducing apoptosis and inhibiting the proliferation of ovarian cancer cells. Inhibition effect was obviously related with the time of collecting serum and the route of administration.
