重肌灵片在重症肌无力小鼠模型T细胞活化中的作用

摘    要:AchR免疫C57BL/6小鼠建立EAMG小鼠模型,观察重肌灵片在自身免疫性重症肌无力小鼠T细胞活化过程中的作用。结果显示重肌灵片抑制AChR诱导的EAMG小鼠淋巴细胞增殖、DTH反应并降低血清中抗AChR抗体;抑制CD28和CTLA-4的表达、增强CD80和CD86的表达,但对CD40和CD40L的表达无显著影响;并降低Th细胞中表达IFN-γ的阳性细胞数、升高表达IL-4的阳性细胞数。提示重肌灵片显著抑制EAMG小鼠T细胞的活化,其抑制作用可能是通过调节细胞表面协同刺激分子的表达,及抑制Th细胞向Th1极化、促进其向Th2极化实现的。

The effect of a Chinese medicine Zhongjiling tablet on the activation of T lymphocytes in experimental autoimmune myasthenia gravis mouse model

Abstract:The effect of a Chinese medicine Zhongjiling tablet of the activation of T lymphocytes in experimental autoimmune myasthenia gravis(EAMG) of mouse was investigated, in which the EAMG mouse model was established by means of immunization with acetylcholine receptor(AchR), and the T lymphocyte proliferation and DTH reaction were inspected. The expression of co-stimulatory molecules and the percentage of Th1/Th2 cells were assayed by flow cytometry. It was demonstrated that Zhongjiling tabled could suppress the T lymphocyte proliferation and DTH reaction induced by AchR and decrease the production of the anti-AchR antibody. In addition it could decrease the expressions of CD28 and CTLA-4 on the surface of lymphocytes, and increase the expressions of CD80 and CD86 on lymph node cells. However, no difference on the expressions of CD40 and CD40L could be demonstrated between the EAMG mouse model and the control group and Zhongjiling showed no any effect on them, but it could reduce the percentage of the IFN-γ-secreting cells and increase the IL-4-secreting cells. It is significant that the mechanism by which the Chinese medicine Zhongjiling tablet suppresses the T lymphocyte activation in EAMG mouse model might be due to its regulatory effect on the expression of costimulatorg molecules so as to suppress the polarization towards Th1 cells and to facilitate that towards Th2 cells.
