富血小板血浆对骨髓间充质干细胞RLA表达的影响

摘    要:目的 探讨富血小板血浆(platelet-rich plasma,PRP)对骨髓间充质干细胞(bone mesenchymal stem cells,BMSCs)免疫原性的影响. 方法 体外培养BMSCs后,将体积分数5%和10%的PRP与BMSCs共培养,应用real time-PCR检测体积分数5%和10% PRP对BMSCs的主要组织相容性抗原RLA Ⅰ类基因及RLAⅡ类基因表达的影响,流式细胞仪检测PRP对RLA-DR表达的影响,单向混合淋巴细胞反应(MLR)的方法检测PRP对兔外周血淋巴细胞(peripheral blood lymphocyte,PBL)增殖反应的影响. 结果 与对照组BMSCs相比,real time-PCR结果显示,5%和10% PRP组BMSCs RLA Ⅰ类基因和RLAⅡ类基因表达差异无统计学意义(P>0.05);流式细胞术结果显示,5%和10% PRP组BMSCs RLA-DR表达差异无统计学意义(P>0.05);MTS试验结果显示,5%和10% PRP组BMSCs PBL增殖差异无统计学意义(P>0.05). 结论 PRP对BMSCs主要组织相容性抗原RLA Ⅰ类基因及RLAⅡ类基因表达无显著性影响.

Abstract:Objective To investigate the effect of platelet-rich plasma(PRP) on the immunogenicity of bone mesenchymal stem cells (BMSCs).Methods After BMSCs were cultured with 5% and 10% PRP in vitro,real-time PCR was used to examine the effects of PRP on RLA Ⅰ and RLA Ⅱ expression,flow cytometry was used to examine the effect of PRP on RLA-DR expression,and one way mixed lymphocytic reaction (MLR) assay was used to examine the effect of PRP on variation of peripheral blood mononuclear cell(PBMC).Results Compared with negative control group,the expression of RLA Ⅰ,RLA Ⅱ and RLA-DR in BMSCs was not significantly different by real-time PCR and flow cytometry assay in 5% and 10% PRP groups (P > 0.05),and the proliferation of PBL was also not different in BMSCs by MTS assay(P > O.05).Conclusion The results suggest that PRP may have no significant effect on expression of RLA Ⅰ and RLA Ⅱ in BMSCs.
