MDR1基因多态性与肝癌易感性的关系

摘    要:目的:探讨MDR1基因位点单核苷酸多态性与肝癌易感性的关系。方法:采用病例对照研究,应用聚合酶链反应限制性片段长度多态性(PCR RFLP)基因型分析技术测定原发性肝癌组及正常健康对照组各100例MDR1基因G2677T/A位点。以χ2检验比较MDR1多态基因型及相关危险在病例与对照之间分布的差异,采用非条件Logistic回归分析多态基因型与肝癌发生危险度的关系。结果:肝癌组MDR1基因2677A携带型低于健康对照组(<0.05);经Logistic回归分析发现2677A携带型的个体患肝癌的危险显著降低,2677A携带型暴露于危险因素下患肝癌风险降低。肝癌组MDR1基因2677T携带型高于健康对照组(<0.05);经Logistic回归分析发现2677T携带型的个体患肝癌的危险增加,2677T携带型暴露于危险因素下患肝癌风险增高。结论:MDR1基因2677A多态性可能是防止肝癌形成的保护因素,2677T多态性可能是肝癌形成的易感因素。

Abstract:Objective:To study the association between genetic polymorphisms of MDR1 and hepatic cell cancer susceptibility. Methods:The experimental subjects composed of 100 cases with primary hepatocellular carcinoma and 100 healthy controls. The polymorphism genetic type of G2677T/A of MDRI was analyzed by PCR-RFLP. The distribution differences of polymorphisms of MDR1 and related risk between the case group and control group were analyzed by chi-square test. The relationship of polymorphism genotype with risk of hepatocellular carcinoma was analyzed by logistic regression. Results : It was observed by logistic regression analysis that the risk of suffering hepatoeellular carcinoma reduced significantly in individuals carrying at least one allele of A, the risk of suffering hepatocellular carcinoma increased significantly in individuals carrying at least one allele of T. Conclusion: MDR1 gene polymorphism of 2677A may be a protective gene of hepatoeellular carcinoma, 2677T may be a risk gene for hepatocellular carcinoma.
