银翘散对流感病毒感染小鼠模型肺组织病毒载量和IFN-γ mRNA表达的影响

摘    要: 目的研究银翘散对流感病毒载量和IFN-γ mRNA表达的影响。方法除正常对照组小鼠不进行处理外,其他小鼠经甲型流感病毒感染后分为模型对照组,银翘散大、中、小剂量组。使用Real-time RT-PCR方法分别在流感病毒感染后第1,2,3,5,7天检测小鼠肺组织病毒载量和IFN-γ mRNA表达。结果银翘散可以降低肺组织病毒载量,其中大剂量组在各时间点均可降低病毒载量(P<0.05,P<0.01),中、小剂量组在第3天和第5天均可降低病毒载量(P<0.01,P<0.05);银翘散可以提高肺组织IFN-γ mRNA表达,其中大剂量组在各时间点均可提高IFN-γ mRNA表达(P<0.01,P<0.05),中剂量组在第2,3,5天可以提高IFN-γ mRNA表达(P<0.01),小剂量组在第3,5天可以提高IFN-γ mRNA表达(P<0.01)。结论银翘散可能通过上调流感病毒感染小鼠肺组织内IFN-γmRNA相对表达量,来发挥抗流感病毒的作用。

Abstract:OBJECTIVE To investigate the effects of Yinqiaosan on influenza virus load and IFN-γ mRNA expression.METHODS The mice were divided into control group,influenza virus group,Yinqiaosan high dose group,Yinqiaosan middle dose group and Yinqiaosan low dose group.Real-time PCR technique was used to detect virus load and IFN-γ mRNA expression in mice lung at the 1st day,2nd day,3rd day,5th day and 7th day after being infected.RESULTS The high dose of Yinqiaosan decreased virus load at every time point(P<0.05,P<0.01).The middle and low dose decreased virus load at the 3rd day and 5th day(P<0.01,P<0.05).Lung IFN-γ mRNA expression was increased in high dose group at every time point(P<0.01,P<0.05),IFN-γ mRNA expression in middle dose group was increased at the 2nd day,3rd day and 5th day(P<0.01),Yinqiaosan in low dose group can increase IFN-γ mRNA expression was increased in low dose group at 3rd and 5th day(P<0.01).CONCLUSION Yinqiaosan exerts antiviral effects against influenza virus by upregulating expression of IFN-γ mRNA.
