低毫安(10mA)电化学疗法对体外肝癌细胞凋亡的诱导及对细胞周期的影响

摘    要:目的:探讨低毫安(10 mA)电化学疗法对体外肝癌细胞凋亡的诱导及对细胞周期的影响。材料和方法:将等量肿瘤细胞置于细胞培养板.按随机原则分为对照组和治疗组,治疗组按电量不同又分为四组:3C、5C、10C、20C(C为电量库仑)电化学治疗后6小时和24小时用流式细胞仪分别检测各组细胞周期变化及凋亡率。结果:低毫安(10mA)电化学治疗后6小时检测结果:肝癌细胞凋亡率及细胞周期无明显改变。治疗后24小时检测结果:治疗组凋亡率明显增高(P<0.05),各期细胞比例随电量的增加处于G_0-G_1期的比例有逐渐增高趋势,而S期细胞比例有下降趋势。结论:低毫安电化学治疗在6小时内对肝癌细胞凋亡率及对细胞周期的影响不明显,24小时后凋亡率及细胞周期变化明显。

Abstract:Purpose: To study the effect of Electrochemical Therapy (EChT) with low milli-Ampere (10mA) on apoplosis and cell cycle of liver cancer cells. Materials and Methods: Equal number of liver cancer cells was put into cell cultured plate. They were divided into control group and treated group by random. Treated group was subdivided into 4 groups according to the different charge: 3 C. 5 C, 10C. 20 C (C: coulomb). 6 hours and 24 hours after treatment, the rate of apoptosis and the percentage of cell cycle were detected by flow cytometry. Results: 6 hours after treatment, the rate of apoptosis and the percentage ol different cell cycle phases did not change obviously. 24 hours alter treatment, the rale of apoptosis in treated groups increased obviously (P<0.05); the percentage of G0-G1 phase had an increasing tendency and the percentage of S phase hail a decreasing tendency as coulomb increasing. Conclusions :Within 6 hours of low mA EchT, the rate of apoptosis and the percentage of cell cycle did not change obviously, but 24 hours after EchT changed obviously.
